With the advent of sophisticated techniques of genetic engineering, we can now readily purify and isolate DNA. Name and explain the different steps involved in the separation and isolation of DNA fragments once cut by restriction endonucleases.

Gel electrophoresis
Negatively charged DNA fragments (produced by restriction endonuclease) move towards anode through agarose gel, smaller fragments move further, separated fragments are stained with ethidium bromide, followed by exposure to UV radiation, extraction of DNA bands by elution.
