• Since DNA is a hydrophilic molecule it cannot pass through cell
membrane.
• Calcium increases the efficiency with which DNA enters the
bacterium through pores in its cell wall

Biolistic guns or gene guns are used to bombard rDNA coated on gold or tungsten particles with high velocity into host (plants) cells.
